Quantification of Mefenamic acid by ultraviolet spectrophotometer in tablet dosage form

BACKGROUND OF MEFENAMIC ACID

Mefenamic Acid is N-(2,3-xylyl) anthranilic acid, chemical class of Mefenamic acid is NSAID and is member of anthranilic acid derivatives.

To determine the amount of any chemical either in its pure or dosage form, the need of assay comes into picture, by using appropriate procedure.

Few ailments like pain, arthritic pain, migraine related to menstruation are treated by the help of mefenamic acid. The principle of ultraviolet spectroscopy and Beer lamberts law are the basis chosen to determine the amount of mefenamic acid in tablet dosage form.1 Other dosage forms can be analysed subjected to scientific optimization adapted in the procedure.2

REQUIREMENTS

  • Mefenamic acid (API)
  • 1,4-dioxane
  • Meff-p tablets (or any other brand)
  • 50.0 ml volumetric flasks
  • Mortar Pestle
  • 0.45 µ filter paper
  • UV Spectrophotometer

PROCEDURE

Standard solution

Add 10.0 mg of pure active pharmaceutical ingredient (API) of Mefenamic Acid in 50.0 ml of volumetric flask containing about 20.0 ml of 1,4-dioxane, shake to mix well, make up the volume with 1,4-dioxane .

Thus concentration of solution obtained is 10mg/50ml, i.e. 0.2mg/ml.

Sample solution

Triturate 20 tablets of meff-p (market brand containing 100.0 mg of Mefenamic acid/tablet) to powder in mortar pestle. Now weight the triturated powder equivalent to 10.0 mg of Mefenamic acid, add in 50.0 ml volumetric flask containing about 30.0 ml of 1,4-dioxane. Shake vigorously for about 15 minutes, make up the volume up to the mark. Filter about 10.0 ml of sample solution by 0.45 µ filter paper.

Instrumentation

Before start, correct the baseline and other parameters if any, with blank in both the cuvettes at 353nm. Then take out one cuvette (from 1st slot) and pour standard solution, take at least three readings.

Similarly take at least three readings for sample solution.
