BACKGROUND OF DANAZOL
Danazol, chemically is a synthetic steroid used to treat primarily endometriosis. Besides this, Danazol is also used for symptoms like immune thrombocytopenic purpura, hereditary angioedema. In present procedure simple Ultraviolet method applied to determine the quantity of Danazol in capules.1
The molecular formula and molar mass.is C22H27NO2 and 337.5 g/mol respectively. Due to its significant side effects the drug is generally not prescribed for more than six months.
REQUIREMENTS
- Danazol API
- Danazol capsules
- 100 ml volumetric flasks
- Chloroform
- 0.45 µm filter paper
- Ultraviolet Spectrophotometer
- Glass Bealker/ Suitable glass container
PROCEDURE
Standard solution
Weigh accurately 10 mg of Danazol API in 100 ml volumetric flask containing about 60 ml of chloroform, mix well by gentle shaking and make up the volume up to 100ml by chloroform.
Sample solution
Open and mix the contents of 20 no of Danazol capsules (market brand) in suitable glass container. Weigh and add capsule content powder equivalent (considering lable claim) to 10 mg of Danazol in 100 ml volumetric flask containing about 50 ml of chloroform, shake vigorously and make up the volume up to the mark by chloroform. Filter sample solution by 0.45 µm filter paper.
Instrumentation
Before starting readings
Stable the baseline without standard or sample solution in cuvette with cover closed. Auto zero the instrument by taking chloroform in both reference and sample cuvettes. Take standard solution in cuvette (1st slot) and take three readings at 285 nm by taking standard solution each time from volumetric flask. Clean the cuvette by chloroform. Take three reading for filtered sample solution at 285 nm by taking sample solution each time from volumetric flask.
