Experiences I gained

Hot spring are the areas where the groundwater is heated geothermally arising from earth crust as a result emergence of spring occurs, this definition I choose just to clarify what is a hotspring on which my project work is based upon, these hotspring carry a biota which are known as thermophiles; they are known to occur at such higher temperatures by increasing their suphur bonds as a result they can withstand a temperature of 90°C. Therefore, I have collected my sample from a hotspring in Odisha known as “Atri”, the water sample has been collected in sterile sealed cap bottles and bought to my laboratory which is in Odisha. The samples are immediately plated to revive the microorganism at a very fresh period and then incubated overnight. Then a repeated culturing of samples were carried out to study the highest zone of hydrolysis, then a series of experiment was done to report the growth curve of the organism having highest zone of hydrolysis. Gram staining of the organism showed the type and shape of organism which also underwent many bioparametric characterization viz., temperature, pH, antibiotic assay etc, and finally three strains were further extended for protein purification and enzymatic assay. This protein purification had to be carried in a different lab, which gave lot of panic to our daily schedule. Then, enzymatic assay and protein estimation are totally a separate experiment for which a person need to learn at the earliest that is before starting any protein work. The ammonium precipitation method is followed by dialysis which is a work of two days, in between of this, person gets time to prepare for next day’s work schedule. After a series of cold room in-out experiences, I got my results having level better enzymatic activity that has to be achieved. Repeated failures with a determined mind and proper time management helped to show me the way towards successful completion of my research.

My work had several limitations some can be discussed that is working in different laboratories for such type of works you need a lot of patience and time management skills. Another one is funding which really affects the research work in a country like India. Still all these hurdles couldn’t stop me to reach my destination of microorganism showing higher l-asparaginase (enzymatic) activity.
