These days a significant progress has been made in the path of blood-brain permeation. A lot of researches are made to know about the molecular properties that helps in the determination of brain permeation. These researches can help a lot in breaking the blood brain barrier which are a nothing but a network of vessels forming structural and chemical barrier between the brain and the blood circulation system. This network of vessel ate endothelial cell that lacks pores, thus forming a block for circulation of blood. discovering blood brain barrier permeability, will be a novel path for neurotherapeutic drugs. As it almost impossible to determine experimentally the blood brain barrier, therefore researchers are doing it basing on various computational models. Recently, AA Toporav Et al, developed a qualitative structure-activity relationship model (QSAR) for blood brain barrier (BBB) permeability, on 159 set of compounds who have BBB permeability data. It is known as Monte Carlo technique that involves corelating weights of various molecular structures. The predictive model was developed by selecting correlation weights of various molecular features. The research study mainly includes methods like k-nearest neighbours and support vector machine approaches.

The complete research included all those important descriptors that highly affects the BBB permeability. The development and study of these models and different factors affecting the permeability helps in estimation of BBB permeability of drug candidates at initial stage of drug development.
