BACKGROUND
Hanging drop method is a process of observing the motility in bacteria and is a wet mount method. The method is used in dark illumination and gives a very clarified view of the microbial motility.1 The objective of this experiment is to study the motility of live bacteria for studying properties of active locomotion.

REQUIREMENTS
Apparatus: Glass slides with depression in the middle
Inoculating loop
Microscope
Bunsen burner
Paraffin wax
Coverslip
Motile bacteria fresh culture
Vaseline
Toothpick

PROCEDURE
Take a clean slide and a cover slip. To the edges of the cover slip apply some Vaseline with the help of toothpick. In the centre of the coverslip keep a loopful of the broth culture.2 Now put the clean slide upside down over the cover slip. After the coverslip attaches to the slide, turn the slide so that the cover slip remains above the slide, so that the drop is seen hanging over from the cover slip to the slide. Now keep the prepared slide under the microscopic slide holder and align it in such a way that drop edge is in the lower objective. Observe the motility through the eyepiece and slightly and slowly raise the objective by the use of coarse adjustment until the drop edge is seen as an irregular line that crosses the field. Adjust the light using diaphragm. Now observe the cells for their morphology as well as their true motility.

CONCLUSION
By this motility test we can easily differentiate between bacteria that are motile and that are non-motile.2 This is an important step for identification of bacteria on the basis of different properties.
