Protein denaturation by egg albumin method (in vitro)

BACKGROUND

Protein denaturation is considered to be one of the reasons for inflammation. NSAIDs prevent protein denaturation along with COX enzyme inhibition.1 

The aim of this experimental work is to study in vitro anti-inflammatory activity by protein denaturation (egg albumin method).

REQUIREMENTS

Drug: Diclofenac sodium

Chemicals: Egg albumin, Phosphate-buffered saline (pH 6.4), Distilled water

PROCEDURE

Mix of 0.2 ml of egg albumin (collected from fresh hen’s egg), phosphate-buffered saline (PBS, pH 6.4) 2.8 ml and different concentrations (10, 20, 30, 40, 50 µg/ml) of Diclofenac sodium (2 ml each). Prepare a same volume with double-distilled water as control. Incubate the mixtures at 37°C in BOD incubator for 15 min and heat at 70°C for 5 min. Cool the solutions and measure absorbance at 660 nm. Take the vehicle as blank.2  Calculate percentage of inhibition of denatured protein by following formula.

CONCLUSION

From this experiment we can conclude, tested drug inhibited denaturation of protein and showed anti inflammatory activity.
