Preparation and Staining of Bone Marrow Smear

BACKGROUND

The aim of the experiment is preparation and staining of bone marrow smear. The semi-solid tissue found in the spongy or cancellous portions of bones is called as bone marrow. The contents are hemopoietic cells, marrow adipose tissue and supportive stromal cells. It constitutes of 4% of the body mass of the human being. The structure of bone marrow is a mixture of cellular and non-cellular components.

The progenitor cells mature into blood and lymphoid cells. The hemopoietic stem cells produce white blood cells, red blood cells and platelets. The bone marrow stroma has fibroblasts, macrophages, adipocytes, osteoblasts, osteoclasts, endothelial cells.1

REQUIREMENTS

Apparatus:      Bone marrow aspiration needle

10 ml plastic syringe

PROCEDURE

The slide is placed in a slanting position in a cup. The bone marrow aspirate of about 0.2 ml is discharged into the elevated part of the slide. The rest of the contents are placed in a screw cap jar containing about 2 ounces of 10% formaldehyde solution.

On the slanted slide, the bone marrow particles gets separated which are picked and smeared on another slide. The remaining particles are washed into specimen bottle by pouring the formalin bone-marrow mixture over the slide. The bone marrow smear is stained.

The bone marrow particles are placed in Templefiber bag in a small funnel. The jar rinsed with formalin is poured into the bag. The erythrocytes pass through, but the marrow particles are left behind. The bag is enclosed in a tissue capsule and processed with autotechnicon. The sections of bone marrow are made and stained with hematoxylin, eosin and Gomori’s method for iron.2

The wright’s or Wright-Giemsa stains are the most commonly used stains for bone marrow aspirate smears for bone marrow studies.

The preparation and staining of bone marrow helps in diagnosing diseases of bone marrow.
