Drug/chemical analysis

Why pH is considered as a critical parameter in analysis by HPLC?

In HPLC analysis pH of mobile phase to be considered as the critical parameter.

The pH of mobile phase affects compound peak shape, retention time and to the stationary phase also.

The pKa of the drug to be taken into consideration while deciding the pH of the mobile phase. If pka of drug and mobile phase pH is same: then peak for drug shows splitting as for ionized and unionized form. The pH of mobile phase should differ from pKa of the drug by 2 units.

The pH of mobile phase to be kept in between 2-8 as silica stationary phase columns dissolve above particular pH.

Modified stationary phases also available for commercially.
