BACKGROUND

Thermal death point (TDP) of bacteria is basically the study of effect of heat on the growth of bacteria. In simple terms it is the time needed to kill bacteria in a medium of liquid culture at a particular temperature.

So, the basic objective of this test is to determine the thermal death point of a given bacterial sample.

REQUIREMENTS

Sample:      Bacterial sample

Others:       Nutrient Agar plates

    Inoculating loops

    Incubator

PROCEDURE

Take two nutrient agar plate and divide it into five quadrants. On each quadrant assign time like 0.15 sec, 2 min, 5 min, 15 min. This time will depict at particular temperature for how much time the organism was heated.1 Now take your sample culture that were heated in different temperatures. Under proper aseptic conditions, with the help of an inoculating loop streak the culture on each part of the quadrant.2 When you are finished incubate the plates at 37 degrees centigrade.

CONCLUSION

Based on the growth of the bacteria the thermal death point can be estimated.3 The quadrant in which there is no growth is the temperature and the time required to destroy the bacteria.
