BACKGROUND
IMViC test is performed for identification of bacteria after doing gram staining. These are one of the stable reactions which gives more accurate results than other tests. The experiment comprises the test for studying indole production by microorganism and its motility.

In MR-VP test MR is for production of glucose, fructose and galactose while VP test is done for glucose production but with different end product. The last test is citrate test which identifies the production of citrate. Thus, the basic objective of this experiment is to test the bacteria for studying its various properties that will help in its identification process (IMViC reactions).
REQUIREMENTS
Media: MRVP broth, SIM deep, Simmon citrate agar slants, TTC agar deep
Reagents: Methyl red reagent, Kovac reagent, Barritts reagent A and B
Miscellaneous: Inoculation loop, Bunsen burner
PROCEDURE
Indole test
Take SIM media and inoculate the bacterium with the help of innoculation needle. Then incubate it for 24 hours at 370C.1
Methyl red and Voges Proskauer test
Take MRVP broth and inoculate bacterium in the broth with the help of inoculation needle. Mix it and incubate at 370C for about 24 hours.
Citrate test
In Simmon Citrate agar slants, streak bacterium with the help of inoculation loop. Incubate this slant at 370C for about 24 hours.
Observation
In SIM agar, watch for black precipitation for H2S production and the stab for motility. After that add two-three drops of kovac reagent. If pink colour forms then it is indole positive.1
To the MR-VP broth, for MR test add about 8 drops of methyl-red and shake well, it will react of positive. Similarly, for VP test add, barritts reagent A and B, to the broth. Intense pink colour if developed it indicates positive.
In citrate test, if slant colour changes to royal blue it is citrate positive.
CONCLUSION
With this test (IMViC reactions) we can easily study various properties of bacteria, that will help in its identification.
