Title of research / project:	Isolation, characterization and purification of l-asparaginase producing microorganism from hotspring

Experiences I gained

Thermostable microorganism have gained a higher index in human population as they provide broad spectrum of antibiotic unanimously, to which I opted as my project work. We have collected water and sediment samples for isolation of microorganism from Odisha’s hot spring Taptapani, in sterile collecting tubes, water has been cultured in nutrient agar and Sabouraud dextrose agar to check for the presence of fungus in the sample.

All these wok of mine was carried in our laboratory of college. The microorganism obtained has been checked for its growth phase and temperature resistance in a media containing l-asparagine, gram’s staining has been carried out for characterization of microorganism. After getting the result based on shape and gram positive / negative stain of organism, a continuous protein purification work has been carried out starting with centrifugation at temperature of 4 °C till dialysis of sample each step follows with the enzymatic assay and protein estimation. This work of mine showed that the water sample gave higher enzymatic activity than the sediment sample.

But a final success, which can bring laureals in the biodata of a science student is -constant effort and endurance to face any failure in the midway of work, so that success will make noise by achieving 4 P’s (passion, planning, persistence and purpose) of life for any work.

After winding up of my thesis and viva-voice, an experience I learnt is to carry out an experiment in a specified time with suggestion and experiences of seniors without any physical help more precisely as all are not masters in practicals, so I experienced to isolate an organism lastly in a course of three months completely by using swabbing, assay and centrifugation.

But a final success, which can bring laureals in the biodata of a science student is -constant effort and endurance to face any failure in the midway of work, so that success will make noise by achieving 4 P’s (passion, planning, persistence and purpose) of life for any work. From all paper research and from my work I can conclude that microorganism’s are life saviour and the water samples gave me my achievements, as they showed higher enzymatic activity which can further analyse may be in future to produce more l-asparaginase enzyme which can be further analysed to produce a medicinal compound which can kill cancerous cells.
